Biological Background: CD38 Function and Localization

  • Catalyzes the synthesis of cyclic ADP-ribose (cADPR) from NAD(+), a second messenger critical for glucose-induced insulin secretion. Related references: PMID:7961800, PMID:8253715
  • Synthesizes nicotinate-adenine dinucleotide phosphate (NAADP(+)), a potent Ca(2+) mobilizing second messenger, preferentially from 2'-phospho-cADPR at both pH 5.0 and pH 7.4. Related references: PMID:16690024
  • Functions as a receptor for CD31 on endothelial cells, promoting lymphocyte activation, proliferation, and transendothelial migration. Related references: PMID:9551996
  • Regulates crucial dendritic cell functions, including CCL21-driven migration, survival, and Th1-polarizing activity. Related references: PMID:16293598
  • In lamina propria T lymphocytes, CD38/CD31 interactions trigger a signaling cascade involving LCK and LAT, leading to cytokine release. Related references: PMID:11259373
  • Highly expressed in pancreas, liver, kidney, brain, testis, ovary, placenta, and malignant lymphoma/neuroblastoma (UniProt tissue specificity).
  • Localized at the cell surface as a single-pass type II transmembrane glycoprotein, featuring disulfide bonds and N-linked glycosylation (UniProt keywords: Signal-anchor, Disulfide bond, Glycoprotein).
  • Undergoes alternative splicing, producing multiple isoforms (UniProt keyword: Alternative splicing).

Experimental Guidance and Technical Tips

  • The immunogen spans amino acids 43–300 of human CD38, covering the extracellular domain; this design supports detection of cell surface CD38 in flow cytometry (FC) and immunohistochemistry (IHC-P).
  • For western blot (WB), expect bands at approximately 34 kDa (full-length) and 14 kDa (potentially a processed or splice variant). Validate with appropriate positive controls, such as pancreatic or lymphoid cell lines.
  • In IHC-P applications, consider optimizing antigen retrieval (e.g., citrate pH 6.0 or EDTA pH 9.0) and antibody dilution using known positive tissues like tonsil or kidney.
  • The antibody is confirmed to react with human and cynomolgus monkey CD38. For other species, conduct species-specific validation.
  • For ELISA, the antibody may serve as either a capture or detection reagent; pairing with a suitable detection system should be optimized experimentally.
