Biological Background: CD38 Function and Localization
- CD38, also known as ADP-ribosyl cyclase/cyclic ADP-ribose hydrolase 1, is a multifunctional transmembrane glycoprotein that belongs to the ADP-ribosyl cyclase family.
- It enzymatically synthesizes cyclic ADP-ribose (cADPR), a second messenger involved in glucose-induced insulin secretion, and also has cADPR hydrolase activity. Related references: PMID:7961800, PMID:8253715
- CD38 converts NADP+ to nicotinate-adenine dinucleotide phosphate (NAADP+), a potent Ca2+ mobilizer, with substrate preference depending on pH. Related references: PMID:16690024
- As a receptor, CD38 binds to CD31 on endothelial cells, promoting lymphocyte activation, proliferation, and transendothelial migration. Related references: PMID:9551996
- In dendritic cells, CD38 regulates maturation-dependent functions including CCL21-driven migration, survival, and Th1 polarization. Related references: PMID:16293598
- In lamina propria T lymphocytes, CD38/CD31 engagement triggers LCK and LAT activation, leading to cytokine release. Related references: PMID:11259373
- CD38 is localized to the cell surface membrane and is expressed at high levels in pancreas, liver, kidney, brain, testis, ovary, placenta, malignant lymphoma, and neuroblastoma.
- The protein undergoes alternative splicing, producing isoforms with approximate molecular weights of 13 kDa and 34 kDa (UniProt: P28907).
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 43–300 of human CD38, covering a large extracellular region; this design may help detect both soluble and membrane-bound forms.
- This product is validated for flow cytometry (FC). For other applications, consider titration and sample-specific optimization.
- Given CD38 expression in multiple tissues, include appropriate positive and negative controls (e.g., lymphoma cell lines) in FC experiments.
- Store the antibody as recommended (ice bag shipping) to maintain conjugate stability; protect from light during handling.
