Biological Background: CD36 Function and Localization
- CD36 (also known as Platelet glycoprotein 4, FAT, GPIV, Scarb3) is a multifunctional transmembrane glycoprotein receptor belonging to the class B scavenger receptor family.
- It binds a broad spectrum of ligands including thrombospondin, oxidized LDL, long-chain fatty acids, collagen, and microbial diacylated lipopeptides, triggering receptor clustering and downstream signaling (PubMed:18332148, PubMed:7685021).

- The immunogen corresponds to amino acids 30–439 of mouse CD36, covering a large portion of the extracellular domain; this region shares high homology with rat CD36, consistent with the observed cross-reactivity.
- In Western blot, a principal band around 53 kDa is predicted; however, glycosylation and other modifications may result in higher apparent molecular weights. Use reducing conditions and appropriate protein markers.
- For IF/ICC and IHC-P, CD36 immunoreactivity is expected at the plasma membrane and possibly Golgi; include relevant controls such as macrophage-rich tissues or known positive cell lines.
- Given its polyclonal nature, this antibody may be used to capture CD36 in ELISA; for sandwich applications, consider pairing with a monoclonal CD36 antibody.
- Validate specificity in your sample system, especially when working with complex tissues, by including knockout or knockdown controls.
