Biological Background: CD35/CR1 Function and Localization

  • CD35/CR1, also known as complement receptor type 1 or C3b/C4b receptor, is a large membrane glycoprotein of approximately 224 kDa that belongs to the regulators of complement activation (RCA) family (SWISS: P17927).
  • It functions as an immune adherence receptor, mediating the capture and clearance of complement-opsonized pathogens and immune complexes by erythrocytes and phagocytes (PubMed:2963069). Related references: PMID:2963069
  • CD35 inhibits spontaneous complement activation by accelerating the decay of C3/C5 convertases of both alternative and classical pathways, and serves as a cofactor for factor I-mediated cleavage of C3b and C4b (PubMed:2972794, PubMed:8175757). Related references: PMID:2972794 PMID:8175757
  • Upon ligand binding, CD35 contributes to the generation of regulatory T cells from activated helper T cells, highlighting its role in immune regulation (PubMed:25742728). Related references: PMID:25742728
  • It also acts as a receptor for Epstein-Barr virus, facilitating viral entry into host cells.
  • CD35 is primarily localized on the plasma membrane and is expressed on erythrocytes, a subset of T cells, mature B cells, follicular dendritic cells, monocytes, and granulocytes.
  • The protein is characterized by multiple short consensus repeat (SCR) domains (also known as sushi domains), extensive glycosylation, and disulfide bonds, and it carries the Knops blood group antigen system.
  • Additional keywords: innate immunity, host-virus interaction, signal peptide, transmembrane helix.

Experimental Guidance and Technical Tips

  • For Western blot, consider using whole cell lysates from human granulocytes or CD35-transfected cells; the expected band is around 224 kDa, and the polyclonal antibody recognizes an epitope within amino acids 420–550.
  • For immunofluorescence/ICC, validate fixation and permeabilization conditions, as CD35 is a membrane protein; staining should localize to the cell surface.
  • For ELISA, the antibody may be used for antigen capture or detection; optimal coating conditions and antibody concentration should be determined empirically.
