Biological Background: CD35/CR1 Function and Localization

  • Complement receptor type 1 (CR1, CD35; gene name CR1) is a membrane immune adherence receptor that mediates the binding and clearance of complement-opsonized pathogens and immune complexes by erythrocytes and monocytes/macrophages. Related references: PMID:2963069
  • CR1 inhibits spontaneous complement activation by impairing the formation and function of the alternative and classical pathway C3/C5 convertases, and by serving as a cofactor for factor I-mediated cleavage of C3b to iC3b, C3c, C3d,g, and of C4b to C4c and C4d. Related references: PMID:2972794, PMID:8175757
  • Upon ligand binding, CR1 contributes to the generation of regulatory T cells from activated helper T cells, playing a role in immune regulation. Related references: PMID:25742728
  • CR1 acts as a receptor for Epstein-Barr virus, facilitating viral entry.
  • CR1 is a transmembrane protein localized on the plasma membrane.
  • CR1 is expressed on erythrocytes, a subset of T cells, mature B cells, follicular dendritic cells, monocytes, and granulocytes.
  • CR1 belongs to the regulators of complement activation (RCA) family and is characterized by multiple sushi (complement control protein) domains.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the C-terminal intracellular region (amino acids 1940–2039) of human CR1. Consider epitope accessibility in membrane preparations or whole-cell staining, and note that glycosylation or protein interactions may influence binding.
  • For immunohistochemistry (IHC-P and mIHC) with paraffin-embedded sections, standard antigen retrieval methods are recommended. Validate staining in tissues with known CR1 expression, such as kidney, spleen, or tonsil.
  • In immunofluorescence (IF-P), consider permeabilization conditions given the large molecular weight (224 kDa) and transmembrane nature of CR1. Include appropriate controls.
  • For ELISA, the peptide immunogen approach enables sensitive detection; however, performance should be verified with the specific sample matrix and recombinant protein standards.
  • As a rabbit monoclonal antibody, use anti-rabbit IgG secondary reagents for detection. For multiplex IHC, confirm compatibility with other primary antibodies and detection systems.
