Biological Background: CD35 Function and Localization

  • Complement receptor type 1 (CR1), also known as the C3b/C4b receptor, is a membrane glycoprotein encoded by the CR1 gene (Gene ID: 1378).
  • It serves as an immune adherence receptor on erythrocytes and phagocytes, capturing complement-opsonized pathogens and immune complexes for clearance from the circulation. Related references: PMID:2963069
  • CR1 negatively regulates complement activation by impairing C3/C5 convertase formation and functioning as a cofactor for factor I-mediated proteolysis of C3b and C4b. Related references: PMID:2972794 PMID:8175757
  • Ligand engagement of CR1 promotes the generation of regulatory T cells from activated helper T cells, highlighting its immunomodulatory role. Related references: PMID:25742728
  • The receptor is expressed on erythrocytes, a subset of T cells, mature B cells, follicular dendritic cells, monocytes, and granulocytes.
  • CR1 belongs to the regulators of complement activation (RCA) family and features multiple sushi domains, disulfide bonds, and glycosylation.
  • It also acts as a receptor for Epstein-Barr virus and carries the Knops blood group antigen system.

Experimental Guidance and Technical Tips

  • The immunogen spans amino acids 901-1095 of human CD35, covering a region within the extracellular domain involved in ligand binding; consider this when interpreting reactivity in functional assays.
  • For Western blotting, a band at approximately 224 kDa is expected; use extended transfer times and low-percentage gels to ensure high-molecular-weight protein resolution.
  • When performing IHC-P or IF/ICC, validate staining patterns in tissues or cells known to express CD35, such as tonsil, spleen, or peripheral blood mononuclear cells.
  • For ELISA, optimize coating conditions and antibody dilutions using purified CD35 protein or complement-coated complexes as targets.
  • Confirm compatibility with your sample preparation and detection systems, as cross-reactivity beyond human has not been tested.
