Biological Background: CD34 Function and Localization

• CD34 is a hematopoietic progenitor cell antigen encoded by the CD34 gene (P28906).
• It functions as a possible adhesion molecule with a role in early hematopoiesis, mediating the attachment of stem cells to the bone marrow extracellular matrix or stromal cells.
• CD34 may act as a scaffold for the attachment of lineage-specific glycans, presenting carbohydrate ligands to selectins, thereby facilitating stem cell binding to lectins expressed by stromal cells or other marrow components.
• The protein is a single-pass type I membrane protein, localizing to the cell surface (Membrane).
• It is selectively expressed on hematopoietic progenitor cells and the small vessel endothelium of a variety of tissues.
• Post-translational modifications include extensive glycosylation and phosphorylation; alternative splicing results in multiple isoforms with observed molecular weights of 35–41 kDa.

Experimental Guidance and Technical Tips

• For Western blotting, consider reducing conditions and note that glycosylation may produce multiple bands in the 35–41 kDa range; validate detection using a relevant cell lysate.
• In immunohistochemistry (IHC-P), antigen retrieval may be necessary; include positive controls such as bone marrow or endothelial-rich tissues.
• For immunocytochemistry (IF/ICC), optimize permeabilization and confirm staining with appropriate controls.
• This antibody, raised against the extracellular domain (aa 32-290), is suitable for detecting both membrane-bound and shed forms of CD34.
