Biological Background: CD34 Function and Localization
- CD34 (also known as GIG3, MORT1) is a hematopoietic progenitor cell antigen encoded by the CD34 gene.
- It functions as a possible adhesion molecule with a role in early hematopoiesis, mediating attachment of stem cells to the bone marrow extracellular matrix or directly to stromal cells.
- Acts as a scaffold for lineage-specific glycans, allowing stem cells to bind to lectins expressed by stromal cells or other marrow components; presents carbohydrate ligands to selectins.
- Subcellularly, CD34 is a membrane protein with a single transmembrane domain.
- It is selectively expressed on hematopoietic progenitor cells and the small vessel endothelium of a variety of tissues.
- Classified under keywords: alternative splicing, cell adhesion, glycoprotein, membrane, phosphoprotein, and signal.
- The full-length protein has a calculated molecular weight of approximately 40.7 kDa; the product data lists 41 kDa.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the intracellular portion of CD34; thus the antibody likely detects total CD34 regardless of glycosylation state.
- For Western blotting, a band at approximately 41 kDa is expected; however, due to heavy glycosylation, mature CD34 often migrates at 90–120 kDa; validate migration in your sample.
- In immunohistochemistry (IHC-P), consider using heat-induced antigen retrieval for optimal staining; the antibody is suitable for paraffin-embedded sections.
- For multiplex IHC (mIHC), ensure compatibility of this rabbit monoclonal with other primary antibodies from different host species.
- Cross-reactivity with mouse and rat has been reported; validate in the relevant tissue or cell line.
