Biological Background: CD34 Function and Localization
- CD34, known as hematopoietic progenitor cell antigen, is a cell surface glycoprotein that functions as an adhesion molecule in early hematopoiesis, facilitating stem cell attachment to bone marrow extracellular matrix or stromal cells. It also acts as a scaffold for lineage-specific glycans and presents carbohydrate ligands to selectins.
- The protein is localized to the plasma membrane, consistent with a single-pass type I transmembrane topology.
- Multiple isoforms are generated through alternative splicing, contributing to functional diversity.
- CD34 is post-translationally modified by glycosylation and phosphorylation, which may regulate its adhesive properties.
- Tissue specificity: CD34 is highly expressed in hematopoietic progenitor cell lines, brain, and testis, with moderate expression in thymus, spleen, and bone marrow. It is not detected in adult liver. In kidney, expression is limited to the thin limb of Henle's loop. Related references: PMID:31605441 PMID:1709048
Experimental Guidance and Technical Tips
- The immunogen, a recombinant fusion protein spanning amino acids 35–287, includes the majority of the extracellular domain, making this antibody well-suited for detecting surface CD34 in IF/ICC and flow cytometry assays.
- For flow cytometry, consider titrating the antibody to determine the optimal concentration for your cell system and perform appropriate isotype controls.
- In IF/ICC experiments, validate fixation and permeabilization conditions, as epitope accessibility may be influenced by membrane protein localization.
- As a monoclonal antibody, expect consistent lot-to-lot performance, which is advantageous for longitudinal studies and assay standardization.
