Biological Background: CD337/NKp30 Function and Localization

  • CD337/NKp30, also known as natural cytotoxicity triggering receptor 3 (NCR3), activating natural killer receptor p30, or NKp30, is a cell membrane receptor encoded by the NCR3 gene.
  • It is selectively expressed on the surface of resting and activated natural killer (NK) cells, with weak expression observed in the spleen.
  • The receptor plays a critical role in NK cell-mediated cytotoxicity by recognizing extracellular ligands such as BAG6 and NCR3LG1.
  • Upon ligand engagement, NKp30 triggers NK cell activation, leading to targeted killing of tumor cells and other abnormal cells.
  • Beyond direct cytotoxicity, NKp30 engagement promotes myeloid dendritic cell (DC) maturation by eliminating immature DCs and inducing NK cells to secrete TNF-α and IFN-γ.
  • Structurally, NKp30 is a transmembrane glycoprotein containing a disulfide bond and an immunoglobulin-like domain, with a molecular weight of approximately 21.6 kDa.
  • The protein undergoes alternative splicing, yielding isoforms with distinct biological properties, and its 3D structure has been elucidated.
