Biological Background: CD328 Function and Localization
- Sialic acid-binding Ig-like lectin 7 (Siglec-7), also known as CD328, is a putative adhesion molecule that preferentially binds to alpha-2,3- and alpha-2,6-linked sialic acids and disialogangliosides. The sialic acid recognition site may be masked by cis interactions on the same cell surface.
- Functions as an inhibitory receptor in immune responses: upon ligand-induced tyrosine phosphorylation, it recruits cytoplasmic phosphatases via SH2 domains, blocking signal transduction through dephosphorylation of signaling molecules.
- Mediates inhibition of natural killer (NK) cell cytotoxicity, playing a key role in regulating innate immunity.
- May also be involved in hematopoiesis; in vitro studies show it inhibits differentiation of CD34+ cell precursors toward the myelomonocytic lineage and suppresses proliferation of leukemic myeloid cells.
- Expressed predominantly on resting and activated NK cells, and at lower levels on granulocytes and monocytes. High expression is found in placenta, liver, lung, spleen, and peripheral blood leukocytes.
- Localized to the cell membrane as a single-pass type I membrane protein.
- Contains immunoglobulin-like domains, disulfide bonds, and glycosylation sites; classified under the immunoglobulin superfamily and lectin family.

- For Western blotting, a band around 51 kDa is expected in human NK cell lysates (e.g., NK-92 cell line) or tissues like spleen and lung.
