Biological Background: CD31/PECAM1 Function and Localization
- Essential cell adhesion molecule for leukocyte transendothelial migration (TEM); Tyr-690 phosphorylation is required for LBRC trafficking and TEM efficiency. Related references: PMID:17580308 PMID:19342684
- Mediates endothelial cell-cell adhesion through trans-homophilic binding and facilitates neutrophil TEM via heterophilic interaction with CD177. Related references: PMID:27958302 PMID:17580308
- Regulates phagocytosis: homophilic ligation prevents viable leukocyte clearance, while apoptotic leukocyte tethering promotes phagocytosis. Related references: PMID:12110892

- Does not protect against apoptosis.

- The immunogen corresponds to the C-terminal cytoplasmic region (amino acids 630–738) of human CD31/PECAM1, a domain conserved in mouse, supporting the reported cross-reactivity.
- For Western blotting, the predicted molecular weight is ~83 kDa; note that the mature glycosylated form may migrate at ~130 kDa in some cell types.
- For immunofluorescence on paraffin-embedded sections (IF-P) and immunohistochemistry (IHC-P), optimal antigen retrieval conditions should be determined empirically.
- Validate the antibody in the relevant sample system (e.g., HUVECs or leukocyte cell lines) due to tissue-specific isoform expression.
