Biological Background: CD31/PECAM1 Function and Localization
- CD31/PECAM1 is a cell adhesion molecule essential for leukocyte transendothelial migration (TEM) under inflammatory conditions; phosphorylation at Tyr-690 is critical for this process. Related references: PMID:17580308, PMID:19342684
- It mediates homophilic interactions at endothelial cell junctions, contributing to vascular integrity. Related references: PMID:27958302
- PECAM1 regulates phagocytosis: it delivers a detachment signal to prevent phagocytosis of viable leukocytes while tethering and promoting clearance of apoptotic cells. Related references: PMID:12110892
- It modulates bradykinin receptor BDKRB2 activation and downstream ERK1/2 signaling in endothelial cells. Related references: PMID:18672896
- PECAM1 is expressed on platelets, leukocytes, and at endothelial cell borders. Multiple isoforms exist: Long form predominates; Delta12 in trachea; Delta14-15 in lung; Delta14 in heart; Delta15 in brain, testis, ovary, etc. Related references: PMID:18388311, PMID:21464369, PMID:12433657
- The protein contains immunoglobulin domains, disulfide bonds, and undergoes post-translational modifications such as glycosylation, palmitoylation, and phosphorylation.
- Subcellular localization includes the cell membrane, membrane rafts, and cell junctions.
Experimental Guidance and Technical Tips
- The immunogen covers the cytoplasmic tail (aa 630–738); the antibody may detect both full-length and cleaved fragments under certain conditions.
- For Western blot, a band at approximately 83 kDa is expected in human, mouse, or rat endothelial cell lysates; validate in the relevant sample system.
- In immunofluorescence and immunohistochemistry, membrane and junctional staining patterns are anticipated; optimize fixation and permeabilization protocols for your experimental setup.
- Consider using this antibody for ELISA applications; confirm appropriate coating and detection conditions in your assay.
