- Tyr-679 phosphorylation in the cytoplasmic tail regulates trafficking to the lateral border recycling compartment, essential for directed migration of leukocytes across endothelial junctions.

- For western blotting, the predicted molecular weight is approximately 81 kDa, but extensive N‑linked glycosylation can cause migration at 120–130 kDa. Use endothelial cell lysates (e.g., HUVEC, bEnd.3) or platelet extracts as positive controls.
- In immunofluorescence (IF-P) and immunohistochemistry (IHC-P), PECAM1 exhibits membranous and junctional staining. For IHC-P, antigen retrieval may be required; test citrate or EDTA buffer and compare with isotype controls.
