Biological Background: BST2 Function and Localization
- BST2 (bone marrow stromal antigen 2), also known as tetherin or HM1.24, is an interferon-induced antiviral host restriction factor encoded by the BST2 gene in humans.
- It efficiently blocks the release of diverse enveloped viruses by tethering nascent virions to the cell membrane, preventing their cell-free spread. This activity has been demonstrated against HIV-1, HIV-2, SIVs, EIAV, FIV, HTLV-1, HCV, Ebola virus, Marburg virus, Lassa virus, SARS-CoV, and many others. - Two isoforms exist due to alternative initiation: isoform 1 and isoform 2. Both function as viral restriction factors but differ in signaling activity and sensitivity to HIV-1 Vpu-mediated degradation. - BST2 localizes to the cell membrane (including lipid rafts), the trans-Golgi network, late endosomes, and the apical cell membrane in polarized epithelial cells.

- The protein is predominantly expressed in liver, lung, heart, and placenta, with lower levels in pancreas, kidney, skeletal muscle, and brain. It is highly expressed during B-cell development and on T-cells, monocytes, NK cells, and dendritic cells. Overexpression is observed in multiple myeloma cells.
- Key post-translational modifications include a GPI-anchor, disulfide bonds, and N-linked glycosylation. It also features a coiled-coil domain and a transmembrane helix.
