Biological Background: CD317/Bst2 Function and Localization
- CD317, also known as bone marrow stromal antigen 2 (BST-2), HM1.24 antigen, or DAMP-1, is an interferon-inducible host restriction factor that blocks the release of diverse enveloped viruses by tethering nascent virions to the cell membrane.
- It acts as a physical tether, linking virions to the cell surface and to each other, leading to their internalization and degradation or retention, thereby limiting cell-free viral spread.
- Targets include retroviruses (HIV-1, MMTV, MLV), filoviruses (Ebola virus), arenaviruses (Lassa virus), and rhabdoviruses (VSV).
- Beyond antiviral activity, CD317 inhibits cell surface proteolytic activity of MMP14, reducing MMP15 activation and suppressing cell growth and migration.
- It provides negative feedback on IFN production by plasmacytoid dendritic cells through LILRA4/ILT7 signaling.
- Subcellular localization: Golgi apparatus, trans-Golgi network, cell membrane, late endosomes, membrane rafts, and apical cell membrane; found both at the cell surface and intracellular compartments.
- In naive mice, CD317 protein is specifically expressed on type I interferon-producing cells (plasmacytoid dendritic cells).
