  • CD317, also known as bone marrow stromal antigen 2 (BST-2), HM1.24 antigen, or DAMP-1, is an interferon-inducible host restriction factor that blocks the release of diverse enveloped viruses by tethering nascent virions to the cell membrane.
  • It acts as a physical tether, linking virions to the cell surface and to each other, leading to their internalization and degradation or retention, thereby limiting cell-free viral spread.
  • Targets include retroviruses (HIV-1, MMTV, MLV), filoviruses (Ebola virus), arenaviruses (Lassa virus), and rhabdoviruses (VSV).
  • Beyond antiviral activity, CD317 inhibits cell surface proteolytic activity of MMP14, reducing MMP15 activation and suppressing cell growth and migration.
  • It provides negative feedback on IFN production by plasmacytoid dendritic cells through LILRA4/ILT7 signaling.
