Biological Background: LAIR1/CD305 Function and Localization

  • LAIR1 (leukocyte-associated immunoglobulin-like receptor 1, CD305) is an inhibitory immune receptor belonging to the immunoglobulin superfamily.
  • Functions as a negative regulator of cytolytic activity in NK cells, T-cells, and B-cells by recruiting phosphatases PTPN6 and PTPN11 upon tyrosine phosphorylation.
  • Modulates cytokine production in CD4+ T-cells, downregulating IL-2 and IFN-γ, while inducing TGF-β secretion; also suppresses IL-8, IL-10, and TNF secretion.
  • Downregulates IgG and IgE production in B-cells and attenuates B-cell receptor-mediated calcium mobilization.
  • Induces apoptosis and inhibits proliferation in myeloid leukemia cell lines by blocking NF-κB nuclear translocation and IκBα phosphorylation.
  • Located on the cell membrane; expressed on most peripheral mononuclear cells, including NK cells, T-cells, B-cells, monocytes, and dendritic cells; notably absent from germinal center B-cells.
  • Post-translational modifications include glycosylation and disulfide bond formation; alternative splicing generates multiple isoforms with predicted molecular weights of 23 kDa, 29 kDa, and 31 kDa.
  • Also known as LAIR-1, with a role in adaptive immunity; aberrant expression observed in HIV-1 infected patients’ B-cells and chronic active EBV infection.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the extracellular domain (amino acids 22–163), suggesting suitability for detection of surface LAIR1 in flow cytometry and ELISA-based assays.
  • In western blotting, multiple bands around 23–31 kDa may be observed due to alternative splicing and glycosylation; consider using protease inhibitors and confirm molecular weight using a positive control lysate (e.g., PBMC or NK cell lines).
  • For flow cytometry, use a fluorochrome-conjugated secondary antibody on live, unfixed cells to preserve surface epitope recognition.
  • For ELISA, pair this antibody with a detection format optimized for recombinant LAIR1 or cell lysates; cross-reactivity to other family members should be evaluated in the relevant experimental context.
  • As LAIR1 signaling involves phosphorylation, consider phosphatase inhibitors and appropriate stimulation conditions when studying functional readouts.
