Biological Background: LAIR1/CD305 Function and Localization
- LAIR1 (leukocyte-associated immunoglobulin-like receptor 1, CD305) is an inhibitory immune receptor belonging to the immunoglobulin superfamily.
- Functions as a negative regulator of cytolytic activity in NK cells, T-cells, and B-cells by recruiting phosphatases PTPN6 and PTPN11 upon tyrosine phosphorylation.
- Modulates cytokine production in CD4+ T-cells, downregulating IL-2 and IFN-γ, while inducing TGF-β secretion; also suppresses IL-8, IL-10, and TNF secretion.
- Downregulates IgG and IgE production in B-cells and attenuates B-cell receptor-mediated calcium mobilization.
- Induces apoptosis and inhibits proliferation in myeloid leukemia cell lines by blocking NF-κB nuclear translocation and IκBα phosphorylation.
- Located on the cell membrane; expressed on most peripheral mononuclear cells, including NK cells, T-cells, B-cells, monocytes, and dendritic cells; notably absent from germinal center B-cells.
- Post-translational modifications include glycosylation and disulfide bond formation; alternative splicing generates multiple isoforms with predicted molecular weights of 23 kDa, 29 kDa, and 31 kDa.
- Also known as LAIR-1, with a role in adaptive immunity; aberrant expression observed in HIV-1 infected patients’ B-cells and chronic active EBV infection.
