Biological Background: CD304/Neuropilin-1 Function and Localization
- Neuropilin-1 (also known as A5 protein) is a transmembrane receptor encoded by the Nrp1 gene in mouse, functioning as a co-receptor for semaphorins and members of the vascular endothelial growth factor (VEGF) family.
- It plays essential roles in cardiovascular development, angiogenesis, and neuronal circuit formation, mediating chemorepulsive responses to semaphorin 3A (SEMA3A). Related references: PMID:26503042
- NRP1 recognizes a C-end rule (CendR) motif (R/KXXR/K) on its ligands, promoting cellular internalization and vascular leakage. It binds to VEGF165, VEGFB, and PLGF-2, and co-expression with KDR/VEGFR2 enhances VEGF165 binding and chemotaxis.
- In the nervous system, NRP1 guides motor neuron axon guidance and cell body migration; it is required for the caudal migration of facial motor neurons from rhombomere 4 to rhombomere 6 during embryonic development. Related references: PMID:26503042
- Beyond its cell surface roles, NRP1 localizes to the mitochondrial membrane and is involved in mitochondrial iron transport via interaction with ABCB8/MITOSUR.
