Biological Background: CD301/CLEC10A Function and Localization
- CD301/CLEC10A (C-type lectin domain family 10 member A) is a Ca(2+)-dependent C-type lectin receptor that specifically recognizes N-acetylgalactosamine (GalNAc)-terminated glycans on glycoproteins and glycolipids.
Related references: PMID:15802303 PMID:16998493 PMID:17616966
- It binds to various GalNAc-containing epitopes, including Tn antigen, T antigen, LacdiNAc, core 5 and 6 glycans, and gangliosides GM2 and GD2.
Related references: PMID:15802303 PMID:23507963
- On immature dendritic cells, CD301/CLEC10A interacts with Tn-carrying CD45 on effector T cells, downregulating CD45 phosphatase activity and thereby modulating T cell activation threshold, cytokine production, and proliferation.
Related references: PMID:15802303 PMID:16998493 PMID:22213806
- This interaction promotes tolerogenic dendritic cell maturation and the generation of regulatory CD4+ T cells with immunosuppressive functions.
Related references: PMID:15802303 PMID:16998493 PMID:22213806
- As an endocytic pattern recognition receptor, CD301/CLEC10A internalizes Tn-antigen-containing tumor antigens into endolysosomes for MHC class I and II presentation, contributing to antitumor immunity.
Related references: PMID:15802303 PMID:17616966 PMID:17804752
-
Subcellularly, CD301/CLEC10A localizes to the cell membrane, early endosome membrane, and lysosome membrane.
-
It is expressed in myeloid antigen presenting cells, such as dermal dendritic cells and those in lymph nodes and skin.
-
Relevant keywords include adaptive immunity, innate immunity, glycoprotein, disulfide bond, alternative splicing, and endocytosis.
Experimental Guidance and Technical Tips
- The immunogen (aa 60-180) lies within the extracellular region containing the carbohydrate recognition domain (CRD), which is well-conserved between human and mouse, in line with the observed cross-reactivity.
- For Western blot, the predicted molecular weight is 35 kDa, but glycosylation may result in higher observed bands; use relevant positive controls such as dendritic cell or myeloid cell lysates.
- For ELISA, consider pairing with appropriate secondary antibodies and using coated recombinant CD301/CLEC10A or cell-based assay systems.
- As a polyclonal antibody, lot-to-lot variability should be assessed for quantitative assays; validate specificity via antigen-blocking or knockdown experiments.
