Biological Background: CD30 Function and Localization
- CD30, also known as TNFRSF8 or Ki-1 antigen, is a member of the tumor necrosis factor receptor superfamily. It is encoded by the TNFRSF8 gene (aliases CD30, D1S166E).
- It functions as a receptor for the ligand TNFSF8/CD30L and may regulate cell growth and transformation in activated lymphoblasts. It activates NF-kappa-B to modulate gene expression. Related references: PMID:8391931 PMID:8999898
- Alternative initiation and alternative splicing generate multiple isoforms.
- The protein localizes to the cell membrane and cytoplasm.
- Post-translational modifications include glycosylation, phosphorylation, and disulfide bond formation. It contains a signal peptide, transmembrane helix, and repeat regions.
- CD30 is expressed in alveolar macrophages at the protein level.
- It belongs to the TNF receptor family and has a reference proteome status.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular domain (amino acids 19-379), making this antibody suitable for detecting cell surface CD30 in applications such as flow cytometry, IHC, and live-cell IF/ICC.
- For Western blot, multiple bands around 14 kDa, 50 kDa, and 63 kDa may be observed due to glycosylation and isoform variations. Validate with a positive control lysate from a CD30-expressing cell line (e.g., Hodgkin lymphoma cells).
- Consider performing antigen retrieval (heat-induced epitope retrieval, pH 6 or 9) for IHC-P staining of formalin-fixed paraffin-embedded tissues.
- For IF/ICC, optimize fixation and permeabilization conditions to balance antigen detection and cellular morphology.
- Validate the antibody in the relevant sample system and assay format before routine use, as performance may vary depending on target expression levels and sample preparation.
