Biological Background: CD3 epsilon Function and Localization
- CD3E is the epsilon chain of the T-cell surface glycoprotein CD3, also known as T3E, TCRE, IMD18, or CD3epsilon.
- It is a component of the TCR-CD3 complex on T lymphocytes, playing an essential role in adaptive immunity. Related references: PMID:15294938 PMID:15546002 PMID:2470098
- Upon TCR engagement by antigen-presenting cells, CD3 chains transmit signals across the membrane via ITAM phosphorylation by LCK and FYN, leading to ZAP70 recruitment and downstream signaling. Related references: PMID:2470098 PMID:40592325
- CD3E ITAM phosphorylation creates docking sites for the kinase ZAP70, converting it into an active enzyme.
- CD3E is crucial for correct T-cell development.
- It mediates internalization and down-regulation of TCR-CD3 complexes via endocytosis sequences in its cytosolic region. Related references: PMID:10384095 PMID:26507128
- Additionally, CD3E acts as a receptor for ITPRIPL1, inhibiting T-cell activation by promoting NCK1 interaction and blocking ERK-NFkB signaling. Related references: PMID:12110186 PMID:38614099
- Subcellular localization is at the cell membrane; the protein features a signal peptide, immunoglobulin domain, transmembrane helix, and cytoplasmic ITAMs.
- Keywords: adaptive immunity, disulfide bond, phosphoprotein, receptor. Predicted molecular weight ~23 kDa.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal extracellular region (aa 1-126) of human CD3 epsilon, suggesting the antibody likely targets an extracellular epitope; consider this for flow cytometry (FC) and immunofluorescence (IF-P) applications on live, non-permeabilized cells.
- For western blot (WB), CD3 epsilon migrates at approximately 23 kDa under reducing conditions; validate the observed band size in your sample system using appropriate positive controls (e.g., Jurkat T-cell lysate).
- For immunohistochemistry on paraffin-embedded sections (IHC-P), antigen retrieval steps (heat-induced, pH 6 or 9) should be optimized as the epitope may be masked.
- In ELISA, pair this antibody with a matched detection antibody to establish a sandwich assay, and confirm specificity with recombinant CD3 epsilon protein.
