Biological Background: CD276/B7-H3 Function and Localization
- CD276 antigen, also known as B7-H3, B7H3, B7RP-2, and 4Ig-B7-H3, is a member of the B7 family of immune checkpoint proteins.
- It exists as two isoforms (isoform 1 and isoform 2) that appear redundant in modulating CD4+ T-cell responses; isoform 2 additionally enhances cytotoxic T-cell induction and interferon-gamma production.
- CD276/B7-H3 may regulate T-cell-mediated immune responses and plays a protective role in tumor cells by inhibiting natural killer cell-mediated lysis, serving as a potential marker for neuroblastoma.
- It may be involved in acute and chronic transplant rejection and in regulating lymphocytic activity at mucosal surfaces.
- The protein could play a key role in establishing a suitable immunological environment at the maternal-fetal interface during pregnancy.
- CD276/B7-H3 is a membrane protein (transmembrane) with an extracellular region containing immunoglobulin-like domains, disulfide bonds, and glycosylation sites.
- It is ubiquitously expressed in many tissues but not detectable in peripheral blood lymphocytes or granulocytes; weakly expressed in resting monocytes, and expressed in dendritic cells, extravillous trophoblast cells, and Hofbauer cells of the placenta.
- Keywords from UniProt: Alternative splicing, glycoprotein, phosphoprotein, disulfide bond, transmembrane helix, signal peptide.
Experimental Guidance and Technical Tips
- This antibody was generated against a synthetic peptide spanning amino acids 29-245 of human CD276/B7-H3 (NP_079516.1), covering part of the extracellular domain; it may recognize both isoforms.
- For Western blot, consider testing at a molecular weight of ~57 kDa under reducing conditions in human cell lysates that express CD276/B7-H3 (e.g., certain tumor cell lines or placental tissues).
- In IHC-paraffin applications, validate antigen retrieval methods due to the target’s membrane localization.
- For ELISA, optimize coating and detection conditions to ensure specificity for native or recombinant CD276/B7-H3.
