Biological Background: CD275/ICOS Ligand Function and Localization
- ICOS ligand (also known as B7-H2, B7RP-1, GL50, and LICOS) is a member of the immunoglobulin superfamily.
- It serves as a ligand for the T-cell-specific receptor ICOS, providing costimulatory signals for T-cell proliferation and cytokine secretion. Related references: PMID:10549624 PMID:10617205 PMID:10657606
- ICOS ligand also induces B-cell proliferation and differentiation into plasma cells. Related references: PMID:10549624 PMID:10617205 PMID:10657606
- Subcellularly, the protein localizes to the cell membrane.
- It is expressed in lymphoid tissues, predominantly in B-cell areas of the lymph nodes and spleen, and also detected on resting splenic T-cells (at protein level). Related references: PMID:10549624 PMID:10617205 PMID:10657606
- Additionally, ICOS ligand is found in various non-lymphoid tissues including lung, thymus, kidney, liver, peritoneum, and testis. Related references: PMID:10549624
- The protein contains disulfide bonds and glycosylation sites, as indicated by keywords.
- It may play a role in modulating secondary immune responses and in skewing maternal T-cell cytokine profiles toward a Th2 phenotype during pregnancy.
Experimental Guidance and Technical Tips
- The immunogen, covering the extracellular region (aa 47-270) of human CD275, is likely to generate antibodies recognizing the native protein. Consider using recombinant protein controls to confirm specificity.
- For Western blotting, the predicted molecular weight is approximately 36 kDa; however, glycosylation may cause the observed band to run higher. Optimize blocking conditions and antibody dilution to minimize non-specific binding.
- This antibody cross-reacts with mouse, making it suitable for studies in murine models. Validate detection sensitivity and specificity in your specific sample system.
- For ELISA applications, the antibody can be used as a detection or capture reagent. Titrate the antibody concentration to achieve optimal signal-to-noise ratio and consider pairing with a matched antibody pair for sandwich ELISA formats.
