Biological Background: CD272/BTLA Function and Localization
- CD272/BTLA (B- and T-lymphocyte attenuator) is an inhibitory receptor on lymphocytes that negatively regulates antigen receptor signaling by recruiting the phosphatases PTPN6/SHP-1 and PTPN11/SHP-2. Related references: PMID:12796776, PMID:14652006
- It may interact in cis or in trans with TNFRSF14, with cis interactions maintaining a resting state in naive T cells, while trans interactions can provide survival signals to effector T cells during adaptive immune responses. Related references: PMID:19915044
- CD272/BTLA is a type I transmembrane glycoprotein containing an extracellular immunoglobulin domain and a cytoplasmic tail with immunoreceptor tyrosine-based inhibitory motifs.
- It is primarily expressed on the surface of splenic T and B cells, and also found in lymph nodes, macrophages, NK cells, and dendritic cells, with very low expression in somatic tissues.
- The protein undergoes alternative splicing and post-translational modifications including glycosylation and phosphorylation.
