Biological Background: CD27 Function and Localization

  • CD27 is a costimulatory immune-checkpoint receptor belonging to the tumor necrosis factor receptor superfamily (TNFRSF7), expressed on the surface of T-cells, NK-cells, and B-cells. Related references: PMID:28011863
  • Upon binding to its ligand CD70, CD27 activates the TRAF2-PTPN6 signaling axis that inhibits LCK phosphorylation, promoting T-cell memory formation and transcriptional adaptation. Related references: PMID:38354704
  • The CD70-CD27 pathway is essential for antigen-specific T-cell activation and expansion, which ensures immune surveillance of B-cells. Related references: PMID:28011863
  • CD27 provides negative feedback signaling during leukocyte differentiation, modulating hematopoiesis in immune activation.
  • In adipose tissue, CD27 negatively regulates the function of Th2 lymphocytes, potentially influencing metabolic inflammation.
  • Subcellularly, CD27 localizes to the cell membrane as a single-pass type I transmembrane protein.
  • The protein undergoes post-translational modifications, including N-glycosylation and phosphorylation, and contains three intramolecular disulfide bonds that stabilize its extracellular domain.
  • UniProt annotates CD27 with keywords such as receptor, signal peptide, transmembrane helix, disulfide bond, glycoprotein, phosphoprotein, apoptosis, and disease variant; it is predominantly found in most T-lymphocytes.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide spanning amino acids 1–100 of human CD27, targeting the N-terminal extracellular domain, which is exposed on the cell surface and involved in ligand binding. This design likely generates antibodies that recognize native, surface-expressed CD27.
  • For Western blotting, the predicted molecular weight of CD27 is approximately 29 kDa; however, due to glycosylation, it may migrate as a diffuse band between 30–55 kDa. Validate using a positive control lysate, such as Jurkat or other T-cell lines.
  • This antibody has been tested for WB, IF-P, and ELISA with human and rat species. For immunofluorescence (IF-P), permeabilization is required to detect intracellular epitopes or to verify surface staining. Always include appropriate negative controls.
  • CD27 can be shed from the cell surface upon activation, generating soluble CD27 (sCD27). If detecting sCD27 by ELISA or other methods, consider using cell culture supernatants or serum samples.
  • To ensure optimal performance, store the antibody at -20°C in working aliquots to avoid repeated freeze-thaw cycles. Shipping on ice bags indicates that the antibody is stable under cold transport conditions.
