Biological Background: CD26/DPP4 Function and Localization
- CD26/DPP4 (dipeptidyl peptidase 4) is a multifunctional type II transmembrane glycoprotein and serine exopeptidase, also known as ADABP, ADCP2, DPPIV, and TP103 (UniProt P27487).
- Acts as a costimulatory receptor essential for TCR-mediated T-cell activation, binding ADA, CAV1, CARD11, and PTPRC to induce proliferation and NF-κB activation. Related references: PMID:10900005, PMID:10951221, PMID:11772392, PMID:17287217
- Functions as a serine exopeptidase that cleaves N-terminal dipeptides from polypeptides with penultimate proline, regulating chemokines, growth factors, neuropeptides, and hormones. Related references: PMID:10570924, PMID:10593948, PMID:16254193
- In association with FAP, mediates pericellular proteolysis of the ECM, promoting endothelial cell migration and invasion. Related references: PMID:10593948, PMID:16651416
- Involved in lymphatic endothelial cell adhesion, migration, and tube formation. Related references: PMID:18708048
- Overexpression enhances cell proliferation, a process inhibited by GPC3. Related references: PMID:17549790
- Localizes to the cell membrane, including apical membrane, invadopodium, lamellipodium, and membrane rafts; also secreted.
- Expressed specifically in lymphatic vessels of skin, small intestine, esophagus, ovary, breast, and prostate; also in small intestine crypt and villous cells; very low in colon.
- Serves as a receptor for MERS-CoV-2 coronavirus.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular domain (amino acids 29-766) of human CD26/DPP4. For Western blotting, expect a band near 88 kDa (calculated molecular weight ~88.3 kDa).
- For flow cytometry, ensure cell surface expression of CD26 on activated T cells, as the epitope is extracellular.
- This antibody is reported to react with human samples; cross-reactivity with mouse or rat is not validated. Consider species-specific testing if using other models.
- Validate the antibody in your specific sample matrix and experimental conditions. Endogenous CD26 expression varies by tissue type and activation state.
