Biological Background: CD26/DPP4 Function and Localization
- CD26/DPP4 is a multifunctional type II transmembrane glycoprotein and serine exopeptidase, also known as DPPIV, ADCP2, and TP103.
- Acts as a costimulatory receptor essential for T-cell receptor (TCR)-mediated T-cell activation by binding ADA, CAV1, IGF2R, and PTPRC. Related references: PMID:10900005 PMID:10951221 PMID:11772392
- Functions as a dipeptidyl peptidase IV that removes N-terminal dipeptides from polypeptides with a penultimate proline, regulating chemokines, growth factors, and peptide hormones. Related references: PMID:10570924 PMID:16254193
- In association with fibroblast activation protein (FAP), mediates pericellular proteolysis of the extracellular matrix, promoting endothelial cell migration and invasion. Related references: PMID:10593948 PMID:16651416
- Overexpression of CD26 enhances cell proliferation, a process inhibited by glypican-3 (GPC3). Related references: PMID:17549790
- Serves as a receptor for MERS-CoV (Middle East respiratory syndrome coronavirus) spike protein, mediating viral entry.
- Subcellularly, it is found on the cell membrane, apical membrane of polarized cells, invadopodia, lamellipodia, cell junctions, and membrane rafts; it is also secreted.
- Tissue expression: selectively expressed in lymphatic vessels (not blood vessels) in skin, small intestine, esophagus, ovary, breast, and prostate; also expressed in intestinal crypt and villous cells.
Experimental Guidance and Technical Tips
- Since the immunogen spans the extracellular domain (aa 29-766), the antibody likely targets an epitope accessible in live-cell flow cytometry. Consider titrating antibody concentrations for optimal staining.
- Validate in CD26-expressing cell lines or primary T cells. For intracellular staining, appropriate fixation/permeabilization may be required.
- The antibody may be used for functional studies blocking T-cell co-stimulation or MERS-CoV entry; however, blocking activity should be experimentally confirmed.
- Note that CD26 shed into serum can be captured; thus, avoid serum-containing buffers if surface staining is critical.
