Biological Background: CD262/DR5/TRAILR2 Function and Localization
- Tumor necrosis factor receptor superfamily member 10B (TNFRSF10B), also known as death receptor 5 (DR5) or TRAIL receptor 2 (TRAIL-R2), is a receptor for the cytotoxic ligand TNFSF10/TRAIL. Upon ligand binding, it recruits FADD and caspase-8 to form the death-inducing signaling complex (DISC), leading to caspase-8 activation and initiation of apoptosis. - Alternative splicing generates multiple isoforms; the canonical isoform is a single-pass type I membrane protein.
- Widely expressed in adult and fetal tissues, with very high levels in many tumor cell lines (HeLa S3, K-562, HL-60, SW480, A-549, G-361) and high expression in heart, lymphocytes, liver, pancreas, spleen, thymus, prostate, ovary, uterus, placenta, testis, esophagus, stomach, and intestinal tract; not detectable in brain.
