Biological Background: TRAILR2 Function and Localization

  • TRAILR2, also known as Death receptor 5 (DR5), CD262, or KILLER, is a tumor necrosis factor receptor superfamily member encoded by the TNFRSF10B gene.
  • This receptor specifically binds the cytotoxic ligand TNFSF10/TRAIL. Following activation, it recruits the adapter molecule FADD and caspase-8 to assemble the death-inducing signaling complex (DISC), thereby initiating caspase cascade-mediated apoptosis. Related references: PMID:10549288
  • TRAILR2 also promotes nuclear factor-kappa-B (NF-κB) activation, linking death receptor signaling to inflammatory and immune responses.
  • The protein is essential for endoplasmic reticulum (ER) stress-induced apoptosis, highlighting its role in cellular stress pathways.
  • Widely expressed in adult and fetal tissues, with very high expression in multiple tumor cell lines (HeLaS3, K-562, HL-60, SW480, A-549, G-361). Highly expressed in heart, peripheral blood lymphocytes, liver, pancreas, spleen, thymus, prostate, ovary, uterus, placenta, testis, esophagus, stomach, and throughout the intestinal tract; not detectable in brain.
