Biological Background: CD25 Function and Localization

  • CD25 (also known as p55, IL-2 receptor alpha chain, TAC antigen) is the alpha subunit of the high-affinity interleukin-2 receptor, encoded by the IL2RA gene.
  • The receptor binds interleukin-2 (IL-2) and is critical for the regulation of immune tolerance by modulating the activity of regulatory T cells (TREGs), which suppress the activation and expansion of autoreactive T-cells.
  • CD25 is a transmembrane glycoprotein that contains extracellular Sushi (CCP) domains and a short cytoplasmic tail. It belongs to the type I cytokine receptor family.
  • It is primarily localized on the plasma membrane of activated T cells, but can also be found on regulatory T cells and other immune cells, where it participates in signal transduction upon IL-2 binding.
  • The protein undergoes post-translational modifications including glycosylation and disulfide bond formation, which are important for its structural stability and receptor function.
  • Mutations in IL2RA are associated with immunodeficiency 41 (IMD41) and susceptibility to type 1 diabetes mellitus (IDDM10), highlighting its essential role in immune homeostasis.
  • CD25 is widely used as a marker for Treg cells in immunological studies and is a target for therapeutic antibodies in autoimmune diseases and transplant rejection.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to a region within amino acids 101-200, which is part of the extracellular domain; thus, the antibody is expected to recognize the mature, surface-expressed form of CD25.
  • For Western blot (WB), the predicted molecular weight is approximately 31 kDa, but glycosylation may cause bands to appear at higher molecular weights. Consider using appropriate positive controls (e.g., activated T-cell lysates).
  • For immunohistochemistry (IHC-P) and immunofluorescence (IF-P), paraffin-embedded sections may require antigen retrieval; test in relevant tissue samples known to express CD25, such as lymphoid tissues.
  • In flow cytometry (FC), the antibody may be used to detect membrane-bound CD25 on live cells; ensure appropriate isotype controls and titration.
  • Cross-reactivity with mouse and rat has been reported, but signal intensity may vary; validate the antibody in your specific model system.
  • For ELISA applications, the polyclonal nature may provide enhanced sensitivity; optimize coating and detection conditions.
