Biological Background: CD248 Function and Localization
- Protein identity and isoforms: CD248, also known as endosialin or tumor endothelial marker 1 (TEM1), is a cell surface glycoprotein encoded by the CD248 gene. Alternative splicing produces multiple isoforms.
- Core function in angiogenesis and pericyte biology: CD248 positively modulates PDGF receptor signaling to promote pericyte proliferation, playing a key role in angiogenesis and vascular remodeling. Related references: PMID:20484976
- Coagulation, insulin signaling, and inflammation: Acts as a scaffold for factor X to enhance coagulation activation, modulates insulin receptor autophosphorylation, and regulates LPS-induced proinflammatory cytokine production.

- Subcellular localization: Single-pass transmembrane protein localized to the plasma membrane.

- The immunogen (amino acids 18–150) corresponds to the extracellular region of CD248, which is suitable for generating antibodies that recognize the native surface protein. For Western blot, a band near 81 kDa is expected; however, glycosylation may cause a shift.
- When using this polyclonal antibody, consider validating specificity in your system with appropriate negative controls (e.g., knockout cell lines or tissue) and titrate for optimal dilution in both WB and ELISA. Cross-reactivity with mouse and rat CD248 can facilitate translational studies, but sequence differences should be verified.
