Biological Background: CD244 Function and Localization
- CD244 is a heterophilic receptor of the signaling lymphocytic activation molecule (SLAM) family, with CD48 as its ligand. It modulates immune cell activation and differentiation through homo- or heterotypic cell-cell interactions, linking innate and adaptive immunity.
- CD244 acts as an activating natural killer (NK) cell receptor, stimulating NK cell cytotoxicity, IFN-gamma production, and granule exocytosis. Related references: PMID:10359122, PMID:11714776, PMID:8376943
- Its activating function implicates association with SH2D1A/SAP and FYN, initiating downstream signaling through VAV1, INPP5D/SHIP1, and CBL. Signal attenuation in the absence of SH2D1A involves INPP5D and, to a lesser extent, phosphatases SHP-1 and SHP-2. Related references: PMID:10934222, PMID:15713798
- At early stages of NK cell differentiation, CD244 may function as an inhibitory receptor, potentially ensuring self-tolerance of developing NK cells. Related references: PMID:11917118
- CD244 acts as a costimulatory molecule in NK cells, enhancing signals from other activating receptors such as NCR3 and NCR1. Related references: PMID:10741393
- It is expressed in spleen, peripheral blood leukocytes, lung, liver, testis, and small intestine, and is found on all NK cells, monocytes, basophils, TCR-gamma/delta+ T-cells, and a subset of CD8+ T-cells.
- The protein is a single-pass type I transmembrane receptor located in the cell membrane and membrane rafts. It undergoes alternative splicing and post-translational modifications including glycosylation, disulfide bond formation, and phosphorylation.
Experimental Guidance and Technical Tips
- The immunogen comprises the extracellular domain (amino acids 22–224), which contains the CD48-binding region; this region may be suitable for detecting CD244 in its native conformation.
- For Western blot, anticipate a band around 42 kDa; post-translational modifications (glycosylation) may cause a shift. Validate using a relevant positive control, such as NK cell lysates.
- ELISA can be used to detect CD244 in biological samples; consider optimizing coating conditions and include appropriate negative controls to ensure specificity.
- Cross-reactivity with mouse CD244 indicates potential use in murine models; however, confirm epitope conservation between species and validate in the relevant experimental system.
