Biological Background: CD24 Function and Localization
- CD24, also known as heat stable antigen (HSA), nectadrin, and lymphocyte antigen 52 (Ly-52), is a signal transducer protein expressed on lymphoid, myeloid, and erythroid cells.
- It plays critical roles in cell differentiation, particularly in early thymocyte development, and modulates B-cell activation responses.
- In association with SIGLEC10, CD24 selectively suppresses immune responses to danger-associated molecular patterns (DAMPs) such as HMGB1, HSP70, and HSP90. Related references: PMID:19264983
- CD24 is involved in the control of autoimmunity. Related references: PMID:20200274
- The protein is anchored to the cell membrane via a GPI anchor and has a signal peptide; the mature polypeptide is approximately 8 kDa (7.8 kDa calculated from sequence).
- CD24 is a glycoprotein involved in innate immunity, and its GPI-anchored structure facilitates signaling through release of second messengers.
- It is highly expressed on hematopoietic cells, consistent with its role in immune regulation.
Experimental Guidance and Technical Tips
- For Western blotting, CD24 has a predicted molecular weight of ~8 kDa; use high-percentage gels (e.g., 15% SDS-PAGE) and low-range molecular weight markers for accurate detection.
- In immunofluorescence applications (IF/ICC, IF-P), CD24 localizes to the cell membrane; primary antibody dilution and permeabilization conditions should be optimized for the sample type.
- For flow cytometry, use an anti-rabbit IgG secondary antibody and ensure proper cell surface staining with live, unfixed cells to preserve GPI-anchored epitopes.
- For ELISA, the immunogen sequence (amino acids 27–53) corresponds to the N-terminal extracellular domain of mature CD24; consider using recombinant CD24 as a positive control.
- Since this antibody is rabbit monoclonal and targets mouse CD24, use species-matched secondary antibodies to avoid cross-reactivity with endogenous mouse immunoglobulins in mouse tissue or cell samples.
- Validate antibody specificity using appropriate controls such as CD24 knockout cell lines or blocking peptides, especially when interpreting results from complex tissue lysates.
