Biological Background: CD226/DNAM-1 Function and Localization
- CD226 (DNAM-1) is a cell surface receptor of the immunoglobulin superfamily, critical for intercellular adhesion, lymphocyte signaling, and cytotoxicity mediated by natural killer (NK) cells and cytotoxic T lymphocytes (CTLs). Related references: PMID:8673704, PMID:9712030
- Upon engaging its ligands PVR/CD155 and NECTIN2/CD112 on virus-infected or tumor cells, CD226 acts as a costimulatory receptor, enhancing NK and CTL killing activity. Related references: PMID:26755705, PMID:31253644, PMID:30591568
- Intracellular signaling involves Src kinase-mediated phosphorylation, leading to GRB2 recruitment and activation of VAV1, PI3K, PLCG1, ERK, and AKT pathways, resulting in calcium fluxes and cytotoxic responses. (By similarity)
- CD226 is predominantly expressed on peripheral blood T lymphocytes and NK cells, consistent with its role in immune surveillance.
- The protein is characterized by the presence of immunoglobulin domains, disulfide bonds, and glycosylation, and is subject to phosphorylation as a key regulatory modification.
- It is localized at the cell membrane as a transmembrane receptor.
Experimental Guidance and Technical Tips
- For Western blotting, the predicted molecular weight of CD226 is approximately 39 kDa, but due to glycosylation, bands may appear at higher molecular weights. Validate with appropriate positive controls.
- In ELISA, consider using recombinant CD226 protein as a standard and optimize coating and detection conditions for reliable quantification.
- To ensure specificity, include relevant negative controls, such as lysates from CD226-deficient cells or knockout models.
- As a membrane protein, efficient extraction may require using buffers optimized for transmembrane proteins.
