Biological Background: CD226/DNAM-1 Function and Localization
- CD226, also known as DNAM-1 or PTA1, is a cell surface receptor that plays a critical role in immune intercellular adhesion, lymphocyte signaling, cytotoxicity, and lymphokine secretion by cytotoxic T-cells and NK cells. Related references: PMID:19029379, PMID:24658051, PMID:26552706
- It functions as a costimulatory receptor upon binding to its ligands PVR/CD155 or NECTIN2/CD112 on virus-infected or tumor cells, enhancing the cytotoxic activity of NK cells and CTLs. Related references: PMID:30591568
- Mechanistically, phosphorylation by Src kinases such as LYN or FYN enables binding to the adapter GRB2, leading to activation of VAV1, PI3K, PLCG1, ERK, and AKT, as well as calcium fluxes.
- The protein is localized at the cell membrane and features immunoglobulin domains, a transmembrane helix, disulfide bonds, glycosylation, and phosphorylation sites.
- Also referred to as Platelet and T-cell activation antigen 1, CD226 is involved in platelet and T-cell activation, highlighting its broad role in immune regulation.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular domain (aa 19–254) of mouse CD226/DNAM-1, which may recognize the native receptor on live cells, making it suitable for flow cytometry.
- For FC, titrate the antibody to determine the optimal dilution for your specific cell type and experiment. Consider using appropriate isotype controls.
- As a directly conjugated antibody, no secondary detection reagent is required, reducing background and simplifying multicolor panel design.
- Validate performance in your cell system of interest (e.g., NK cells, T cells), given the reported reactivity with mouse.
