Biological Background: CD226/DNAM-1 Function and Localization

  • CD226, also known as DNAM-1 or PTA1, is a cell surface receptor that plays a critical role in immune intercellular adhesion, lymphocyte signaling, cytotoxicity, and lymphokine secretion by cytotoxic T-cells and NK cells. Related references: PMID:19029379, PMID:24658051, PMID:26552706
  • It functions as a costimulatory receptor upon binding to its ligands PVR/CD155 or NECTIN2/CD112 on virus-infected or tumor cells, enhancing the cytotoxic activity of NK cells and CTLs. Related references: PMID:30591568
  • Mechanistically, phosphorylation by Src kinases such as LYN or FYN enables binding to the adapter GRB2, leading to activation of VAV1, PI3K, PLCG1, ERK, and AKT, as well as calcium fluxes.
  • The protein is localized at the cell membrane and features immunoglobulin domains, a transmembrane helix, disulfide bonds, glycosylation, and phosphorylation sites.
