Biological Background: CD225/IFITM1 Function and Localization

  • CD225/IFITM1 (Interferon-induced transmembrane protein 1) is a 14 kDa transmembrane protein belonging to the dispanin family. It is also known as 9-27, IFI17, LEU13, and DSPA2a.
  • As a potent antiviral restriction factor, IFITM1 inhibits entry of diverse viruses including influenza A, SARS-CoV, SARS-CoV-2, Ebola, Marburg, Dengue, West Nile, HIV-1, and HCV by preventing viral fusion after endocytosis. Related references: PMID:26354436 PMID:33270927
  • It specifically blocks SARS-CoV-2 spike-mediated cell–cell fusion (syncytia formation). Related references: PMID:33051876
  • IFITM1 is implicated in cell adhesion and control of cell growth and migration. Related references: PMID:33270927
  • It plays a key role in the antiproliferative action of IFN-gamma by inhibiting ERK activation or inducing p53-dependent G1 arrest.
  • IFITM1 acts as a positive regulator of osteoblast differentiation.
  • Subcellularly, IFITM1 localizes to the cell membrane and lysosomal membrane.
  • Tissue expression: detected in bone, with markedly elevated levels in colon, cervical, esophageal, ovarian cancers and glioma cell lines.
  • Post-translational modifications include palmitoylation and phosphorylation, annotated under keywords antiviral defense, innate immunity, osteogenesis, and lipoprotein.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 1–80 of human IFITM1; confirm detection of endogenous full-length protein and evaluate potential cross‑reactivity with IFITM2/3 in your specific assay format.
  • For Western blot (expected band ~14 kDa), use high‑percentage Tris‑glycine gels (e.g., 15%) and a PVDF membrane with optimized wet transfer conditions for small proteins.
  • In immunofluorescence and immunohistochemistry, include permeabilization steps and appropriate negative controls (isotype control or IFITM1‑knockout cell/tissue) to validate specificity.
  • When developing ELISA applications, test the antibody as a capture or detection reagent and verify linearity and sensitivity with recombinant IFITM1 protein.
