Biological Background: CD225/IFITM1 Function and Localization

  • Interferon-induced transmembrane protein 1 (IFITM1, also known as CD225, Leu-13, IFI17, DSPA2a) is a member of the dispanin family and is encoded by the IFITM1 gene.
  • Broad-spectrum antiviral restriction factor that inhibits the entry of many enveloped viruses, including influenza A virus, SARS coronaviruses (SARS-CoV and SARS-CoV-2), Ebola virus, HIV-1, and hepatitis C virus. Related references: PMID:26354436 PMID:33270927
  • Mechanistically, IFITM1 blocks viral fusion after endocytosis, trapping virions in endosomes and promoting their lysosomal degradation. In hepatocytes, IFITM1 cooperates with IFITM2 and IFITM3 to restrict HCV entry. Related references: PMID:26354436
  • Also implicated in cell adhesion, growth control, and migration. IFITM1 mediates the antiproliferative effects of IFN-gamma via p53-dependent cell cycle arrest and inhibits SARS-CoV-2 spike protein-mediated syncytia formation. Related references: PMID:33270927 PMID:33051876
  • Localizes primarily to the plasma membrane and lysosome membrane, consistent with its role in blocking viral fusion and directing virions to the degradative pathway.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide from amino acids 50–125, which should generate antibodies recognizing the full-length protein. Validate reactivity with full-length IFITM1 in interferon-stimulated cell lysates.
  • For western blotting, expect a band around 14 kDa. Treatment of cells with type I or type II interferons may upregulate IFITM1 expression.
  • In IHC-P, test on tissues known to express IFITM1 (e.g., bone, tumor sections) and optimize antigen retrieval. Use appropriate controls to confirm specificity.
  • For ELISA, use the immunizing peptide or recombinant IFITM1 as coating antigen; cross-reactivity with other IFITM family members should be checked.
  • Cross-reactivity with mouse allows use in mouse models, but confirm sequence alignment within the immunogen region.
