Biological Background: CD22 Function and Localization
- CD22 is a B-cell-specific inhibitory receptor belonging to the sialic acid-binding immunoglobulin-like lectin (siglec) family. It is most highly expressed on B-lymphocytes and plays a role in B-cell differentiation, antigen presentation, and trafficking to bone marrow.

Phosphorylation of its ITIM domain by LYN kinase recruits the tyrosine phosphatase PTPN6/SHP-1, leading to negative regulation of BCR signaling.

Experimental Guidance and Technical Tips
- The immunogen is a recombinant protein spanning human and mouse CD22, suggesting the antibody recognizes conserved epitopes and is suitable for cross-species detection.
- In Western blot (WB), expect bands between 95–97 kDa. Validate the molecular weight against the appropriate cell lysate (e.g., human/mouse B-cell lines) and confirm specificity using positive and negative controls.
- For immunofluorescence (IF/ICC) and immunohistochemistry (IHC-P), consider optimizing fixation and permeabilization conditions. The antibody may be compatible with both frozen and paraffin-embedded sections.
- When used in ELISA, test the antibody in a standard sandwich or direct format with recombinant CD22 protein or cell-derived samples.
