Biological Background: CD22 Function and Localization
- CD22, also known as Siglec-2 or BL-CAM, is a B-cell-specific sialic acid-binding immunoglobulin-like lectin and a member of the siglec family.
- It is the most highly expressed siglec on B-cells and plays a key role in B-cell differentiation, antigen presentation, and trafficking to bone marrow. Related references: PMID:34330755, PMID:8627166
- CD22 binds alpha 2,6-linked sialic acid residues on surface molecules such as CD22 itself, CD45, and IgM in a cis configuration, and also functions as an adhesion molecule in trans. Related references: PMID:20172905
- It acts as an inhibitory coreceptor on B-cells, negatively regulating B-cell receptor (BCR) signaling by recruiting the tyrosine phosphatase SHP-1 via its ITIM domain, leading to reduced calcium mobilization and cellular activation. Related references: PMID:8627166
- Strong inhibitory signaling through CD22 can induce B-cell apoptosis. Related references: PMID:20516366
- CD22 is localized to the cell membrane and is specifically expressed in B-lymphocytes.
- It is a glycoprotein with multiple immunoglobulin domains and undergoes phosphorylation, alternative splicing, and disulfide bond formation.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the C-terminal intracellular region (aa 708-847) of CD22, containing the ITIM domains. The antibody may recognize intracellular epitopes; therefore, it is suitable for Western blot and immunostaining under permeabilizing conditions.
- For Western blot, expect a band between 95-130 kDa; the predicted molecular weight is ~95 kDa, but glycosylation often results in a higher apparent molecular weight. Validate with appropriate controls.
- For immunofluorescence/ICC, include a permeabilization step to allow antibody access to the intracellular epitope.
- The antibody is reported to cross-react with mouse and rat CD22; verify reactivity in your specific model system.
- ELISA can be employed for detecting CD22 protein or titering applications; optimize assay conditions for your matrix.
