Biological Background: CD22 Function and Localization

  • CD22, also known as SIGLEC2 or BL-CAM, is a member of the sialic acid-binding immunoglobulin-like lectin (siglec) family and is the most highly expressed siglec on B-cells. Related references: PMID:34330755, PMID:8627166
  • CD22 functions as an adhesion molecule by binding alpha 2,6-linked sialic acid residues on itself, CD45, and IgM in cis, and also mediates trans interactions with ligands on other cells. Related references: PMID:20172905
  • As an inhibitory coreceptor, CD22 negatively regulates B-cell receptor signaling by recruiting the phosphatase PTPN6 via its phosphorylated ITIM domain, leading to inhibition of calcium mobilization and cellular activation. Related references: PMID:8627166
  • Upon sufficient negative signaling, CD22 can induce apoptosis in B-cells. Related references: PMID:20516366
  • CD22 is a single-pass type I membrane protein localized to the cell membrane.
  • It is specifically expressed on B-lymphocytes.
  • The protein undergoes post-translational modifications including glycosylation and phosphorylation, and its extracellular region contains immunoglobulin-like domains.
  • Alternative splicing of CD22 generates multiple isoforms.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide from the N-terminal region (aa 1-100); consider using this peptide as a positive control in ELISA to confirm antibody reactivity.
  • For Western blot, a band near 95 kDa is expected; however, due to glycosylation, the observed molecular weight may be slightly higher. Validate in B-cell lysates.
  • This polyclonal antibody may recognize multiple isoforms arising from alternative splicing; assess specificity in your sample system using knockdown or knockout controls if needed.
  • Explore B-cell lines such as Raji or Daudi as positive models for both WB and ELISA applications.
