Biological Background: CD22 Function and Localization
- CD22, also known as SIGLEC2 or BL-CAM, is a member of the sialic acid-binding immunoglobulin-like lectin (siglec) family and is the most highly expressed siglec on B-cells. Related references: PMID:34330755, PMID:8627166
- CD22 functions as an adhesion molecule by binding alpha 2,6-linked sialic acid residues on itself, CD45, and IgM in cis, and also mediates trans interactions with ligands on other cells. Related references: PMID:20172905
- As an inhibitory coreceptor, CD22 negatively regulates B-cell receptor signaling by recruiting the phosphatase PTPN6 via its phosphorylated ITIM domain, leading to inhibition of calcium mobilization and cellular activation. Related references: PMID:8627166
