Biological Background: CD215/IL-15R alpha Function and Localization

  • Interleukin-15 receptor subunit alpha (CD215) is a high-affinity receptor for IL-15, mediating both cis and trans signaling to regulate immune cell homeostasis and activation. Related references: PMID:8530383
  • In neutrophils, CD215 binds and activates the kinase SYK upon IL-15 stimulation, which is required for IL-15-induced phagocytosis. Related references: PMID:15123770
  • Multiple isoforms exist due to alternative splicing; some isoforms do not bind IL-15 and may interfere with signal transduction. Related references: PMID:10480910
  • The receptor localizes to various cellular membranes including the plasma membrane, nucleus membrane, endoplasmic reticulum, Golgi apparatus, and cytoplasmic vesicles; a secreted soluble form (sIL-15RA) is also produced.
  • Expression is detected in neutrophils at the protein level, and in fetal brain with higher levels in hippocampus and cerebellum; soluble forms are abundant in brain structures.
  • Post-translational modifications include N-linked glycosylation, phosphorylation, and disulfide bond formation, as evidenced by UniProt annotation.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 167-267 within the extracellular domain of human CD215/IL-15R alpha; this region may be suitable for detecting both membrane-bound and soluble forms. Consider validating detection of the soluble receptor in your assay system.
  • For Western blot, the predicted molecular weight is approximately 28 kDa. Glycosylation and isoform diversity may lead to additional bands; validate in the relevant sample system with appropriate controls.
  • Immunofluorescence/ICC may reveal membrane and intracellular staining patterns consistent with reported subcellular localizations. Optimization of fixation and permeabilization is recommended.
  • This polyclonal antibody may recognize multiple epitopes; for ELISA, consider using recombinant protein standards to ensure specificity and quantify soluble IL-15RA levels.
