Biological Background: Interleukin-13 receptor subunit alpha-2 Function and Localization
- Interleukin-13 receptor subunit alpha-2 (IL-13Rα2), also known as CD213a2, IL-13R, IL13BP, is a cell surface receptor encoded by the IL13RA2 gene in humans.
- Functions primarily as a decoy receptor that binds IL-13 with high affinity, inhibiting IL-13 and IL-4 signaling through the JAK-STAT pathway, thereby modulating immune responses and inflammation. Related references: PMID:11861389, PMID:17030238
- Additionally, serves as a signaling receptor for IL-13 via an alternative AP-1 pathway, leading to the production of TGFB1, which contributes to fibrotic responses. Related references: PMID:16327802
- Localized to the cell membrane as a single-pass type I transmembrane protein, with an extracellular region containing fibronectin type-III domains and a short cytoplasmic tail.
- Contains a signal peptide, multiple disulfide bonds, and N-linked glycosylation sites, consistent with its role as a membrane glycoprotein.
- Belongs to the type I cytokine receptor family, sharing structural features with other interleukin receptors but lacking the conserved WSXWS motif and intracellular signaling domains required for JAK/STAT activation.
- The UniProt entry (Q14627) lists keywords including 3D-structure, Cell membrane, Disulfide bond, Glycoprotein, Membrane, Proteomics identification, Receptor, Reference proteome, Repeat, Signal, Transmembrane, Transmembrane helix.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular domain (residues 27-355); consider the possible presence of shed soluble receptor when choosing sample types for WB or IP.
- For WB, an expected band near 44 kDa can be observed; pre-incubation with recombinant IL-13 may help confirm antibody specificity.
- Suitable for immunoprecipitation of the native receptor from cell lysates; use appropriate detergents to preserve epitope conformation.
- ELISA applications can be considered for detecting soluble IL-13Rα2 in biological fluids; validate with relevant standard curves.
