Biological Background: CD209 Function and Localization

  • CD209 (DC-SIGN) is a C-type lectin receptor expressed on dendritic cells, alternatively named CLEC4L, CDSIGN, and hDC-SIGN.
  • Functions as a pathogen-recognition receptor that mediates endocytosis of pathogens, leading to antigen presentation to T cells and initiation of adaptive immunity.
  • Serves as a high-affinity receptor for ICAM2 and ICAM3, facilitating dendritic cell rolling, transendothelial migration, and T-cell clustering, thereby promoting T-cell proliferation. Related references: PMID:10721994 PMID:10721995 PMID:11017109

  • In immunofluorescence/ICC, permeabilization may be required to detect intracellular CD209; consider using dendritic cell lines or primary cells as positive controls.
  • For ELISA applications, pairing with a suitable capture/detection antibody is advised; test conditions using recombinant CD209 protein.
  • Given cross-reactivity with mouse and rat, the antibody may be used for studies in rodent models, but validation in the relevant species is essential.
