Biological Background: CD209 Function and Localization
- CD209, also known as DC-SIGN, CLEC4L, or CDSIGN, is a C-type lectin receptor predominantly expressed on the surface of immature dendritic cells.
- It functions as a pathogen-recognition receptor, mediating endocytosis of pathogens and subsequent antigen presentation to T cells via MHC class II molecules, thus linking innate and adaptive immunity.
- As a high-affinity receptor for ICAM2 and ICAM3, CD209 facilitates dendritic cell rolling and transendothelial migration, and forms a first contact between DCs and resting T cells through ICAM3 binding, promoting T cell proliferation. Related references: PMID:10721994 PMID:10721995 PMID:11017109
- CD209 serves as an attachment receptor for numerous viruses, including HIV-1, HIV-2, Ebolavirus, HCV, Dengue virus, SARS-CoV, and Lassa virus, facilitating viral entry into host cells. Related references: PMID:23966408
- In bacterial infections, CD209 recognizes high-mannose N-linked oligosaccharides on pathogens such as Mycobacterium tuberculosis and Helicobacter pylori, contributing to host defense. Related references: PMID:16379498 PMID:16092920 PMID:21203928
- Subcellularly, CD209 localizes primarily to membrane rafts and the cell membrane, but secreted isoforms also exist; it is highly expressed in dendritic cells, placental macrophages, and certain endothelial cells.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide corresponding to amino acids 1–100 near the N-terminus, which suggests the antibody may detect both native and denatured forms, supporting applications such as WB, IHC-P, IF/ICC, and ELISA.
- For Western blotting, a band around 46 kDa is expected; validate with appropriate positive control lysates and consider using a reducing SDS-PAGE gel.
- In IHC-P on formalin-fixed paraffin-embedded tissues, antigen retrieval may improve staining; optimize the retrieval method and antibody dilution for your specific samples.
- For IF/ICC, permeabilization is required to access the intracellular epitope; use a suitable permeabilization buffer and confirm specificity with negative controls.
