Biological Background: CD205/DEC-205 Function and Localization
- CD205, also known as Lymphocyte antigen 75 (LY75), DEC-205, or gp200-MR6, is a type I transmembrane glycoprotein belonging to the C-type lectin domain family 13 (CLEC13B).
- It functions as an endocytic receptor that directs captured antigens from the extracellular space to specialized antigen-processing compartments, playing a key role in antigen presentation.
- CD205 causes reduced proliferation of B-lymphocytes, potentially modulating humoral immune responses.
- The protein contains multiple C-type lectin-like domains (CTLDs) and a fibronectin type II domain, and is heavily glycosylated and phosphorylated.
- CD205 is primarily localized to the membrane and is expressed in spleen, thymus, colon, peripheral blood lymphocytes, as well as myeloid and B-lymphoid cell lines.
- Alternative splicing generates multiple isoforms; isoform 2 and isoform 3 are notably expressed in malignant Hodgkin lymphoma cells (Hodgkin and Reed-Sternberg cells), suggesting a role in cancer biology.
