Biological Background: CD205/DEC-205 Function and Localization

  • CD205, also known as Lymphocyte antigen 75 (LY75), DEC-205, or gp200-MR6, is a type I transmembrane glycoprotein belonging to the C-type lectin domain family 13 (CLEC13B).
  • It functions as an endocytic receptor that directs captured antigens from the extracellular space to specialized antigen-processing compartments, playing a key role in antigen presentation.
  • CD205 causes reduced proliferation of B-lymphocytes, potentially modulating humoral immune responses.
  • The protein contains multiple C-type lectin-like domains (CTLDs) and a fibronectin type II domain, and is heavily glycosylated and phosphorylated.
  • CD205 is primarily localized to the membrane and is expressed in spleen, thymus, colon, peripheral blood lymphocytes, as well as myeloid and B-lymphoid cell lines.
  • Alternative splicing generates multiple isoforms; isoform 2 and isoform 3 are notably expressed in malignant Hodgkin lymphoma cells (Hodgkin and Reed-Sternberg cells), suggesting a role in cancer biology.
  • The full-length protein has a calculated molecular weight of approximately 198 kDa.

Experimental Guidance and Technical Tips

  • The immunogen spans amino acids 216-501, a region within the extracellular domain; consider whether the antibody targets linear or conformational epitopes when optimizing assay conditions.
  • For Western blot, the predicted molecular weight is ~198 kDa; use low-percentage SDS-PAGE gels and extended transfer times to ensure efficient detection of this high-molecular-weight protein.
  • In immunofluorescence/immunocytochemistry, appropriate permeabilization may be required for membrane staining; validate in known CD205-expressing cell lines such as B-lymphoid or myeloid lines.
  • For ELISA, the antibody can be used to detect recombinant CD205 or endogenous protein in cell lysates; pair with appropriate detection reagents.
  • Cross-reactivity is confirmed for human; test cross-reactivity in other species if needed.
