Biological Background: CD204 (MSR1) Function and Localization
- CD204, also known as macrophage scavenger receptor types I and II (MSR1) or scavenger receptor class A member 1 (SCARA1), is a transmembrane glycoprotein that exists in multiple isoforms generated by alternative splicing (Isoform I, II, III).
- It mediates the binding and endocytosis of modified low-density lipoproteins such as acetylated LDL and oxidized LDL, facilitating cholesterol uptake and contributing to foam cell formation during atherogenesis. Related references: PMID:2251254, PMID:34252459
- CD204 recognizes a wide array of pathogen-associated and damage-associated molecular patterns, including bacterial cell wall components and apoptotic cell debris, enabling macrophage clearance functions in innate immunity. Related references: PMID:10722588
- Upon CpG DNA stimulation, CD204 activates LYN kinase, which phosphorylates SYK, leading to recruitment to endosomal TLR9 and enhancement of TLR9 signaling. Related references: PMID:29057947, PMID:40980882
- The protein is primarily localized to the cell membrane, with expression in monocyte-derived macrophages and in tissues such as liver, placenta, and brain.
- Key structural features include a signal-anchor, transmembrane helix, coiled-coil domains, and collagen-like regions; it is glycated and phosphorylated.
- Different isoforms exhibit functional variations; for example, one isoform does not internalize acetylated LDL.
Experimental Guidance and Technical Tips
- For flow cytometry, consider using a live-cell staining protocol to detect surface CD204 expression on macrophages. Optimize antibody titration and include appropriate isotype controls to ensure specificity.
- Since the antibody is rabbit monoclonal, it may provide consistent batch-to-batch performance. Validate the staining in your specific experimental system, especially when analyzing rare cell populations.
- Prepare single-cell suspensions from human PBMCs or tissue-derived macrophages, and use a viability dye to exclude dead cells. The conjugate is compatible with standard flow cytometers equipped with a 561 nm or 594 nm laser.
- CD204 expression can be modulated by certain stimuli; consider testing both resting and activated macrophages to confirm detection across activation states.
