Biological Background: CD1d Function and Localization
- CD1d is an antigen-presenting glycoprotein belonging to the CD1 family of MHC-like molecules, also known as R3G1.
- It binds self and non-self glycolipids and presents them to T-cell receptors on natural killer T-cells, playing a key role in innate immunity.
- Subcellular localization includes the cell membrane, basolateral cell membrane, endosome membrane, lysosome membrane, and endoplasmic reticulum membrane, consistent with its antigen-processing and presentation function.
- Tissue expression is observed on cortical thymocytes, certain T-cell leukemias, and in various other tissues.
- Post-translational modifications include disulfide bond formation and glycosylation, typical of immunoglobulin domain-containing membrane proteins.
- Structural features include a signal peptide, an immunoglobulin domain, a transmembrane helix, and a short cytoplasmic tail.

- For Western blot (WB), CD1d has a predicted molecular weight of ~38 kDa; consider using reducing conditions and validate with a positive control such as human thymocyte lysate.
