Biological Background: CD19 Function and Localization
- CD19, also known as B4 or CVID3, is a B-lymphocyte surface antigen that serves as a coreceptor for the B-cell receptor (BCR) complex, reducing the threshold for antigen-induced activation. Related references: PMID:29523808, PMID:1373518, PMID:16672701, PMID:2463100
- Activation of CD19 triggers phosphatidylinositol 3-kinase (PI3K) signaling and Ca2+ mobilization, crucial for B-cell responses. Related references: PMID:12387743, PMID:16672701, PMID:9317126
- CD19 is dispensable for early B-cell development in the bone marrow but essential for antigen-dependent B-cell differentiation and proliferation in the periphery. Related references: PMID:9317126, PMID:1373518, PMID:2463100
- It is required for normal serum immunoglobulin levels and the generation of high-affinity antibody responses. Related references: PMID:12387743, PMID:16672701, PMID:9317126
- Subcellularly, CD19 is anchored to the plasma membrane and enriched in membrane rafts, where it facilitates signal transduction.
- CD19 expression is observed on marginal zone and germinal center B cells in lymph nodes, as well as on peripheral blood B cells. Related references: PMID:2463100, PMID:16672701
- The protein undergoes alternative splicing, yielding multiple isoforms with potential functional diversity.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide from the C-terminal intracellular region (aa 456–556). For IF-P and IHC-P, optimize permeabilization to expose the intracellular epitope.
- In WB, expect a band near 61 kDa. Include positive controls such as B-cell lines (e.g., Raji, Daudi) or human PBMCs, and validate loading with a housekeeping protein.
- Cross-reactivity is indicated for human only; if using other species, perform preliminary validation.
- For ELISA applications, titer the antibody and establish optimal blocking and detection conditions empirically.
