Biological Background: CD18 Function and Localization
- CD18 (integrin beta-2, ITGB2) is a type I transmembrane glycoprotein that pairs with multiple α subunits (CD11a, CD11b, CD11c, CD11d) to form heterodimeric integrins: LFA-1 (αLβ2), Mac-1 (αMβ2), p150,95 (αXβ2), and αDβ2 (αDβ2).
- These integrins mediate leukocyte adhesion and transmigration by binding ICAMs (1-3), VCAM1, complement iC3b, fibrinogen, and factor X, critical for immune cell trafficking and effector functions (PubMed:1676048, PubMed:23775590, PubMed:38195629, PubMed:29100055, PubMed:10438935, PubMed:8777714, PubMed:9841932, PubMed:11812992, PubMed:28807980, PubMed:18587400). Related references: PMID:1676048, PMID:23775590, PMID:38195629, PMID:29100055, PMID:10438935, PMID:8777714, PMID:9841932, PMID:11812992, PMID:28807980, PMID:18587400
- As a key receptor for apoptotic cell clearance, LFA-1 (αLβ2) cooperates with ICAM3 to promote phagocytosis of apoptotic neutrophils by macrophages (PubMed:23775590). Additionally, αXβ2 recognizes erythrocyte ICAM4 and neuronal ICAM5, linking integrins to erythrophagocytosis and neuron–leukocyte interactions (PubMed:16985175, PubMed:10741396). Related references: PMID:23775590, PMID:16985175, PMID:10741396
- CD18 integrins contribute to natural killer cell cytotoxicity (PubMed:15356110). Mac-1 (αMβ2) is required for CD177‑PRTN3-mediated activation of TNF‑primed neutrophils (PubMed:21193407). Related references: PMID:15356110, PMID:21193407
- Subcellularly, CD18 localizes to the cell membrane and membrane rafts of leukocytes, particularly neutrophils (PubMed:23775590, PubMed:21193407, PubMed:28807980). Related references: PMID:23775590, PMID:21193407, PMID:28807980
- The protein is heavily glycosylated, contains EGF‑like domains, and harbors calcium/magnesium-binding motifs essential for integrin activation. It is post‑translationally modified by phosphorylation and features a pyrrolidone carboxylic acid at its N‑terminus.
- Consistent with its essential role in leukocyte adhesion, ITGB2 mutations are linked to immunodeficiencies such as leukocyte adhesion deficiency (LAD) type I.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N‑terminus (aa 1–100) of human CD18; verify reactivity in the intended species/application via positive control lysates (e.g., human, mouse, or rat leukocytes).
- For Western blot (WB), a predominant band at ~85 kDa is expected; consider using reducing conditions and probing with appropriate secondary antibodies.
- For paraffin immunofluorescence (IF‑P) and immunohistochemistry (IHC‑P), optimize antigen retrieval (heat‑induced epitope retrieval is commonly required) and titrate the primary antibody to reduce background.
- ELISA applications may require coating with the recombinant immunogen or native integrin complexes; pair with a detection secondary to establish a standard curve.
- Due to cross‑reactivity with mouse and rat, validate antibody specificity in the relevant model systems when switching between species.
