Biological Background: CD177 Function and Localization

  • CD177 is a GPI-anchored glycoprotein expressed on a subset of human neutrophils, also known as NB1 glycoprotein or PRV-1.
  • In association with the beta-2 integrin heterodimer ITGAM/CD11b and ITGB2/CD18, CD177 mediates activation of TNF-primed neutrophils, including degranulation and superoxide production. Related references: PMID:21193407
  • By preventing beta-2 integrin internalization and attenuating chemokine signaling, CD177 favors neutrophil adhesion over migration. Related references: PMID:28807980

  • For immunohistochemistry (IHC-P), antigen retrieval conditions may need optimization; because CD177 is expressed on only a subset of neutrophils, staining may be heterogeneous.
  • In ELISA applications, this antibody may be useful as a capture or detection reagent; pairing with a compatible secondary antibody is advised.
  • Cross-reactivity is reported as human; performance in other species should be experimentally validated if needed.
